Usually a Pin of the 2nd IF IC.

I found a schematic of the IC-3210 online. Look at J-10 Pin 5 the SQLS pin. 
That pin will change state when a signal opens the squelch.
I am not sure what voltage level it will be, you can measure that and also 
might have to build a an inverter or have it fire a transistor switch to 
get the voltage you require but it will work for your COS requirement.
