Yes, the hardware has to be connected before the driver will make the entry in /dev.
Because the O.P. had so much trouble typing in Terminal.app here is the exact commands to try:
dkelly at clumsy {121} cd /dev
dkelly at clumsy {122} ls -l tty.usb*
ls: No match.
dkelly at clumsy {123}
I don't have it so "No match."
To further confuse things one could type ls -l /dev/tty.usb*
