Well, I can certainly try and help!

INQUIRY: What is your computer running? Windows XP, Windows
Shitsa^H^H^H^H^H^HVista, Windows 7, Windows 8, or something else?

Now, the question is; can MMTTY hear the audio?

You have two options:
1. If you are running XP; make sure the Stereo Mixer (I think that's
the name of it; I'll confirm tomorrow) is not muted, it's under the
"Recording Devices" menu of the Volume Mixer panel. If that was muted,
well that's your problem; if it isn't, try reversing the mark and
space tones (use the REV button in MMTTY).
2. If you are on any later Windows version... well you're screwed out
of using the Stereo Mixer because MSFT removed that feature (because
of course NO ONE uses that </sarcasm>). So, go get a male-male 3.5mm
cable plug one end into the speaker port, and the other into the
microphone in port. Unfortunately, there goes your computer's audio
output, unless you get a Y-cable as well...
I suggest those two fixes just because getting the ITTY audio to the
program is real problem issue; the remainder are simple "sanity
checks." (Is my shift 170 Hz? Is my mark tone correct? Am I flipped
the right way around? Is my baud rate and charater size correct?...)

Well, you may dislike what I'm about to say, but ditch MMTTY; fldigi
is much, much nicer in my opinion.
Here's the fldigi site: http://www.w1hkj.com/Fldigi.html
It does everything MMTTY does, has a somewhat nicer interface, more
customization; it also does other things (more modes for one... want
to experiment with MT63, Olivia or DominoEX? Go for it.)
Also, if you are using Windows Media Player, please don't VLC is a
significantly less crap one.
Cheers,
Christian
