Yes you will need a different cable for the SL-USB to K3. I'm not
aware of one available commercially, but it's not hard to make one. I
recommend cannibalizing an old twisted pair Ethernet cable. That and a
few connectors (for line in, line out, and PTT on the K3), and you'll
be set. Just remember to match your wiring to the SL's internal
jumpers (or vice versa).
I prefer to use the SL's hardware PTT wired into the K3, rather than
VOX, but it's a matter of preference. Either works fine.
For the MLDX you'll need a serial port in the form of a USB-to-serial
adapter. These have been discussed to death on this reflector, so scan
the archives and you'll get plenty of input on that.
