Check the simple things first. First the microphone gain control that 
its not turned way down or open at some positions. Then the microphone 
connector, followed by the microphone cable at the ends. In my 
experience microphone cables fail most often, more often that tubes and 
at the transmitter end of the cable where solder wicked into the 
stranded wire makes the wire break or the insulation falls off and the 
audio gets shorted. There's always more stress at the ends of the 
microphone cable.

Another test is to enable VOX and see if the relay keys, that will check 
some of the circuits between the microphone and the audio pick off for 
the VOX, but the VOX gain has to be above zero for the VOX to work and 
the antivox can't be too high.

Then check the listed tubes and the diode ring balanced modulator.
