I've used both a FT-8100 and a TM-D700 successfully for cross-band repeat with a Comet dual band antenna. Both have a single SO-239 antenna connector.
I suspect the two vs one antenna question really comes down to where the antenna(s) are mounted. With one antenna, you can probably mount it in an optimal location on the vehicle (see March 2012 QST) but finding a spot for two antennas will probably result in a compromised location for both.
So I'd opt for a single dual-band antenna well located on the vehicle.
OBTW... you can purchase an external VHF/UHF diplexer to split the single transciever antenna connector into separate ports for 2M and 440, but that probably makes more sense for a fixed station setup that has separate gain antennas for each band.
