On a regular Mark 5, at least, if you use a standard serial computer cable, you will need a gender changer at the radio end.  The parameters I use are 4800, no parity, 8 data bits, 2 stop bits, and both DTR and RTS are always on.  Be sure not to use a null modem cable.
